I'd suggest that you first try going into your Defender screen (Virus and threat protection) and disabling Tamper Protection, and then turning off real-time protection. If you don't turn off TP first, turning off real-time protection will be ineffective. But if you go this route, be SURE to go back and change those settings when you're finished.

Theoretically and more than most likely in practice: Defender should pick up on a discrepancy immediately. That’s the one job Defender has, to protect your PC. If a malicious file was able to run, Defender would stop the process. Same answer as above because it’s the same question just worded differently.

Limited Periodic Scanning is intended to offer an additional line of defense to your existing antivirus program’s real-time protection. When Windows 10 Limited Periodic Scanning is turned ON, and even if you are NOT using Microsoft Defender for your real-time protection , the Microsoft Defender user interface and History tab will allow you to view any additional threats that have been detected.
